Diphtheria: A serious bacterial infection that can lead to breathing problems, heart failure, and nerve damage.
Pertussis (whooping cough): A highly contagious respiratory disease, particularly dangerous for infants, causing severe coughing fits that make breathing difficult.
Tetanus: A severe disease caused by bacteria entering the body through wounds, affecting the nervous system and causing painful muscle spasms (lockjaw) and breathing problems.
Hepatitis B: A viral infection that affects the liver and can cause life-threatening infections and severe liver disease, including liver cancer.
Haemophilus influenzae type b (Hib): A bacterium that can cause severe illnesses such as meningitis (infection of the covering around the brain), pneumonia (lung infection), and sepsis (blood poisoning).
Administration and Schedule
The vaccine is administered by intramuscular injection, typically in the thigh of infants. It is generally given in a multi-dose schedule during the first year of life to ensure long-lasting protection. The World Health Organization (WHO) recommends a three-dose schedule, with the first dose as early as 6 weeks of age, followed by subsequent doses at four-week intervals (e.g., 6, 10, and 14 weeks) in many countries.
The DPT-HepB-Hib vaccine can be given at the same time as other routine childhood vaccines, such as polio, measles, and rotavirus vaccines, but at a separate injection site.
Safety and Effectiveness
The pentavalent vaccine has an excellent safety record, with millions of doses administered worldwide. Most side effects are mild and short-term, such as a sore leg where the injection was given, a small lump at the injection site, or a raised temperature. The vaccine is highly effective; for example, the Hib component has reduced cases of Hib infections by over 95% in countries where it has been introduced.
